What it does, and how
Kisspeptin signals through the GPR54 (KISS1R) receptor on hypothalamic GnRH neurons, triggering pulsatile GnRH release that drives downstream LH and FSH from the pituitary, which then stimulates testosterone or estradiol production. The KP-10 fragment retains the receptor-binding C-terminal sequence of full-length kisspeptin while being easier to synthesise. Waljit Dhillo's group at Imperial College London ran the bulk of the human studies, including IV kisspeptin-54 trials in healthy men, hypogonadal patients, and women with hypothalamic amenorrhoea. Effects on libido and emotional limbic activation have been documented in functional MRI studies.
Typical practice
Community practice runs 50 to 200 mcg per day subcutaneous, typically every other day in 4 to 8 week cycles. Some users dose pre-workout or pre-intimate situations for the acute libido effect (peak plasma at ~30 to 60 minutes post-injection). Reconstitution typically 5 mg vial in 2 to 5 mL bacteriostatic water. The published human IV studies used substantially higher per-bolus doses (up to 12 nmol/kg) but those were research single-doses, not chronic stacks. The community sub-q dosing is conservative relative to that and reflects what users do, not what trials validated for chronic use.
The dosing above is community practice, not a regulator-validated protocol. Trial-validated dosing for Kisspeptin-10 in humans does not exist for most use cases listed.
Risks and contraindications
Most-reported side effects: mild flushing post-injection, occasional headache, transient nausea. The HPG-axis stimulation is reversible because kisspeptin acts upstream of the natural pulsatile system; tachyphylaxis (receptor desensitisation) has been observed in continuous-infusion studies, which is why pulsatile or alternate-day dosing is preferred. Women: avoid during pregnancy. Anyone on hormone-affecting medication (TRT, AIs, SERMs, antiandrogens) should consult an endocrinologist before stacking, since the upstream stimulation can cause unpredictable cascades. Cancer history involving hormone-sensitive tumours: hard contraindication.
